A combined live imaging and DESI-Raman approach for dissecting the molecular basis of muscle regeneration

Abstract

Stem cells are critical for regeneration, but this is impaired in ageing. Multiple cues regulate stem cell responses including metabolic and immune signalling. How these cues act to spatially regulate behaviour of stem cells during regeneration and how this affected by ageing is an urgent and important question. Zebrafish telomerase mutants will be used as to understand how ageing affects regeneration by characterising stem cell responses to injury and comparing to metabolite and lipid distribution. Combining live cell imaging, single cell sequencing, Raman imaging and DESI mass spectrometry imaging this project will identify novel ageing-related molecules regulating stem cell function.
