Defining the role of laminin 332 in skin barrier formation

Abstract

Laminin 332 (Lam332) is the most  abundant extracellular matrix protein secreted by epidermal skin cells (keratinocytes) and plays a key role in defining the dermal-epidermal boundary. However, contributions of Lam332 to epidermal homeostasis and cell signaling remain poorly understood.   Our preliminary RNA sequencing data has revealed a novel role forLam332 in regulating keratinocyte lipid synthesis, which in turn plays an important role in formation of the skin barrier.  This project will use biochemistry and advanced microscopy to study in vitro and in vivo skin models and determine how Lam332 signals via adhesion receptors to regulate lipid metabolism. Data arising from this study will provide novel insight into fundamental pathways that control epidermal barrier integrity.
