Mechanistic basis for co-operativity in kinesin-1/cargo recognition

Abstract

Misregulation of cellular transport can contribute to human diseases ranging from neurodegenerative conditions to cancer and even viral infections. Kinesin-1 is one of the most important molecular motors and recently it has become increasingly clear that multiple interactions between the motor and cargo adaptor proteins can regulate its activation. In this project we will employ a structural biology approach (X-ray crystallography and cryo-EM) to study protein complexes between kinesin-1, JIP3, and JIP1 involved in APP transport. Functional hypotheses will be tested taking advantage of a novel in vivo assay to study the biology of ageing Drosophila neurons.
