Abstract
The dynamics of the myosin S1-S2 junction plays a crucial role in muscle contraction, its regulation and heart disease. It is very difficult to characterise this complex, multiprotein complex using traditional approaches. We have therefore embarked on developing a multidisciplinary approach combining computer simulations, NMR spectroscopy, imaging and SAXS. Data from experimental and theoretical methods will allow us to dissect in full spatial and temporal detail how phosphorylation of the regulatory light chain and interactions with myosin binding protein C at the myosin S1-S2 junction are combining to regulate myosin head positioning and thus muscle contraction.
